Accurate mass determination of a bispecific T cell engager (BiTE®) consisting of two single-chain variable fragments (scFvs) recombinantly linked by a nonimmunogenic five-amino-acid chain was analyzed by SEC–MS using a TSKgel® UP-SW3000, 2 µm column under nondenaturing conditions. No signs of particle shedding or sample carryover, which may interfere with MS signal response, were noted in the analysis.

This application describes the use of a 4.6 mm ID × 15 cm TSKgel® UP-SW3000, 2 µm SEC column for fast and accurate mAb aggregate analysis without compromising the quality of the aggregate determination or reproducibility. Results show that the run time of the 15 cm column was completed in 4 min without compromising the resolution of the chromatogram.

In this application note, an ADC was separated using a TSKgel® Butyl-NPR column, the least hydrophobic of the TSKgel HIC columns. Both unconjugated and drug conjugated Trastuzumab samples were successfully separated with baseline resolution. The baseline resolution enabled an easy integration and quantification of different drug pay loads in ADC characterization.
