Webinar Date/Time: Thu, Mar 9, 2023 11:00 AM EST

Samantha Ippoliti from Waters Corporation reviews how to achieve complete and clean trypsin digestion with an enzyme that provides a balance of low levels of missed cleavage, non-specific cleavage, and trypsin autolysis. The applicability of RapiZyme trypsin in both traditional peptide mapping protocols and a variety of alternative workflows will be presented.

Event Overview:

Achieve complete and clean trypsin digestion with an enzyme that provides a balance of low levels of missed cleavage, non-specific cleavage, and trypsin autolysis. This webinar will review how RapiZyme trypsin provides a chemically optimized recombinant trypsin with excellent thermal stability and exquisite levels of autolysis resistance. This method development exercise explores the applicability of RapiZyme trypsin in both traditional peptide mapping protocols and a variety of alternative workflows, including accelerated 30-minute digestions, leisurely overnight digestions, and a one-pot reaction which skips the traditional desalting step.

Key Learning Objectives:

  • How to balance digestion performance parameters during protein digestion method development
  • How to achieve 30-minute protein digestions without sacrificing data cleanliness or digestion completeness
  • Options for eliminating desalting steps without relying on heat-based denaturation
  • How to reduce artificial deamidation in overnight digestions through optimized digestion parameters

Speakers:

Samantha Ippoliti
Senior Scientist
Waters Corporation

Samantha has supported biopharmaceutical characterization and monitoring workflows at Waters Corporation for the past four years; this involved a variety of LC-MS techniques such as intact/subunit and peptide mapping. Before her time at Waters, she spent 5+ years at Novartis in a biologics developability group performing LC-MS assays to support profiling and formulation studies. She graduated with her BS-MS in Chemistry from Northeastern University in 2011.
