Event Overview: The technical issues surrounding the characterization of SARS-CoV-2 spike (S) protein glycosylation and its importance for quality control of expressed spike protein will be discussed. A novel approach based on reversed phase LC-MS/MS with a Q-TOF will be described capable of identifying hundreds of glycopeptides which were used to populate a mass-retention time database. Participants will learn how this database can be incorporated into a simple, powerful, rapid and robust characterization workflow.

  • Challenges of SARS-CoV2 spike protein glycan analysis
  • Novel glycopeptide discovery workflow
  • SARS-CoV2 spike glycopeptide database
  • SARS-CoV2 spike Mass-Retention Time Fingerprinting (MRTF) workflow

Key Learning Objectives:

  • Understanding SARS-CoV-2 spike protein glycan structure
  • Novel approaches to glycopeptide separation & analysis
  • Conceptual understanding of the mass-retention time fingerprinting technique

Speaker: Dr. Rod Chalk, Head of Mass Spectrometry, New Centre for Medicines Discovery, Oxford University
