HDX-MS with online EC reduction enables successful epitope mapping of mAbs to challenging S-S bonded protein targets, e.g., VEGF. The biophysical characterization of VHH-IR4, a BBB-crossing single-domain antibody (sdAb) is described for the first time and was only possible using EC reduction.
